When they first meet, Serena tells Ash that she's a vampire, and since then, they've been traveling together.
In the first movie, he had a relationship with Misty Kyurem. But it ended badly when she was killed by Deino Ash went crazy and tried to kill Serena. However, both of them managed to escape from that place and now they're friends again. In the second movie, he meets another girl named Lucy who seems to like him but then she gets kidnapped by the villain so he has to go on a mission where he has to fight against monsters to save her. At the end of the movie, he realizes that she wasn't really interested in him but rather in his money so she could restore her family's restaurant.
In the third movie, he travels to the new world where he meets two girls named Dawn and Daybreak who seem to like him but then some villains come and attack them so he has to fight them to protect them. At the end of the movie, he realizes that one of them (probably Dawn) loves him too much because she'll never leave him even if he's not human like her.

They even had a real kiss at the end of Season 1, which caused a huge controversy when it was revealed later in the season that she was actually a villain named Dawn. Despite this, they both agreed that they wanted to keep their relationship a secret until after the tournament, so as not to influence the outcome.
After defeating Team Rocket in the final episode of the first season, Serena showed up with everyone else as Ash claimed his first Grand Festival Title. She congratulated him on his victory and they shared an emotional moment before she left. Although she did leave without saying goodbye, this was probably because she knew she would be traveling again soon enough. In any case, this shows that even if Ash didn't know it yet, he has strong feelings for Serena and she for him. This is further confirmed in the sequel series, The Last Wish.

When Ash found out that Serena was hurt, he rushed over to her side only to find out that she had been shot with an arrow by someone named "the hunter".
